目的研究慢性丙型肝炎(CHC)患者外周血CD4+CD25+调节性T细胞与丙型肝炎病毒(HCV)-RNA载量和丙氨酸转氨酶(ALT)活性的相关性。方法流式细胞术计数66例CHC患者和30例健康对照者外周血CD4+CD25+调节性T细胞,实时荧光PCR检测HCV-RNA载量,乳酸脱氢酶法检测ALT浓度。结果 CHC患者外周血CD4+CD25+调节性T细胞显著高于健康对照组(P<0.01),且随着HCV-RNA载量增高,患者外周血中CD4+CD25+调节性T细胞含量逐渐升高(P<0.001),且与HCV-RNA载量取对数后呈正相关(r=0.603,P<0.01),与ALT水平呈正相关(r=0.638,P<0.01)。结论 CHC患者外周血CD4+CD25+调节性T细胞显著升高,与HCV-RNA载量和ALT的升高相关性具有统计学意义。提示CD4+CD25+调节性T细胞可能参与丙型肝炎的进展,为丙型肝炎的诊疗研究提供了新的思路。
Objective To investigate the relationship between CD4 + CD25 + regulatory T cells and hepatitis C virus (HCV) -RNA and ALT in patients with chronic hepatitis C (CHC). Methods CD4 + CD25 + regulatory T cells in peripheral blood of 66 patients with CHC and 30 healthy controls were counted by flow cytometry. HCV-RNA load was measured by real-time fluorescence PCR and ALT concentration by lactate dehydrogenase assay. Results The CD4 + CD25 + regulatory T cells in peripheral blood of patients with CHC were significantly higher than those in healthy controls (P <0.01). The levels of CD4 + CD25 + regulatory T cells in peripheral blood were increased with the increase of HCV-RNA (R = 0.603, P <0.01), and positively correlated with ALT level (r = 0.638, P <0.01). Conclusion The CD4 + CD25 + regulatory T cells in peripheral blood of patients with CHC are significantly increased, which is correlated with the increase of HCV-RNA load and ALT. These results suggest that CD4 + CD25 + regulatory T cells may participate in the progress of hepatitis C, which provides a new idea for the diagnosis and treatment of hepatitis C.
