目的 梯级剂量脂多糖 (LPS)致伤对大鼠全身炎症反应综合征 (SIRS) 肺损伤严重程度的影响。方法 经Wistar大鼠尾静脉注射梯级剂量LPS( 2、4、6、8mg/kg)致伤 ,观察呼吸频率、死亡率 ,检测PaO2 、观察外周血WBC计数及分类大白细胞构成比、观察肺W/D值以及病理学检查。结果 ①LPS致伤 ,可以导致大鼠呼吸困难 ,PaO2 降低 ,外周血WBC计数异常 (LPS小剂量使其升高 ,大剂量使其降低 ) ,外周血大白细胞构成比升高 ,肺组织W /D值升高 ,肺病理改变加重 ;②LPS≥ 6mg/kg致伤 ,上述变化非常显著。结论 ①LPS致伤 ,可以诱发大鼠发生SIRS 肺损伤 ;②LPS 6mg/kg是导致大鼠SIRS 肺损伤失控 ,发生ARDS的临界剂量
Objective To investigate the effect of staged dose lipopolysaccharide (LPS) injury on the severity of lung injury induced by systemic inflammatory response syndrome (SIRS) in rats. Methods Wistar rats were killed by caudal vein injection of cascade dose of LPS (2,4,6,8mg / kg), respiratory rate and mortality were observed, PaO2 was measured, WBC count of peripheral blood and classification of leukocyte were observed, / D value and pathological examination. Results (1) LPS injury caused respiratory dysfunction, decreased PaO2, abnormal WBC count in peripheral blood (increased in low dose of LPS and decreased in large dose), increased leukocyte ratio in peripheral blood and W / D Value increased, pulmonary pathological changes worse; ② LPS ≥ 6mg / kg injury, the above changes were significant. Conclusion ①LPS injury can induce SIRS lung injury in rats. ②LPS 6mg / kg is the critical dose of ARDS which causes uncontrolled SIRS lung injury in rats
