目的优化千里光总生物碱的富集纯化工艺,并分离制备单体生物碱Adonifoline。方法采用正交实验设计法优化千里光总生物碱的富集工艺,采用色谱法分离制备Adonifoline。结果影响千里光总生物碱富集的因素为酸水用量、碱化pH值和氯仿萃取次数。结论优化工艺适合千里光总生物碱的富集,富集后的总生物碱含量与粗提物相比,提高了417倍。色谱法分离制备得到的Adonifoline纯度达到98%以上。
Objective To optimize the enrichment and purification of total alkaloids from Senecio, and to separate and prepare monomeric alkaloids Adonifoline. Methods Orthogonal experimental design was used to optimize the enrichment process of total alkaloids of Senecio ophthalmus. Adonifoline was prepared by chromatographic separation. Results The factors influencing the total alkaloids enrichment were the amount of acid and water, alkalization pH value and times of chloroform extraction. Conclusion The optimized process is suitable for the enrichment of total alkaloids of Senecio, and the content of total alkaloids after the enrichment is 417 times higher than that of the crude extract. The purity of Adonifoline prepared by chromatographic separation was over 98%.
