已知血管平滑肌细胞(VSMC)增殖是动脉粥样硬化、血管再通术后再狭窄等许多血管疾病的基本病理改变之一。VSMC的增殖机理目前尚不清楚,近年大量研究资料证实氧自由基参与动脉粥样硬化的发病过程。但氧自由基与VSMC增殖的关系尚未完全阐明,本实验在培养家兔主动脉VSMC上,观察自由基发生系统(Free radical generating system,FRGS、H_2O_2和酸性环境,pH 6.8)对~3H-胸腺嘧啶核苷(~3H-TdR)和~3H-亮氨酸参入的影响,以阐明自由基及酸中毒对VSMC增殖的关系。
Proliferation of vascular smooth muscle cells (VSMCs) is known to be one of the basic pathological changes of many vascular diseases such as atherosclerosis and restenosis after revascularization. The proliferation mechanism of VSMC is not yet clear. In recent years, a large number of research data confirm that oxygen free radicals participate in the pathogenesis of atherosclerosis. However, the relationship between oxygen free radicals and VSMC proliferation has not yet been fully elucidated. In this study, free radical generating system (FRGS, H 2 O 2 and acidic environment, pH 6.8) The effects of pyrimidine nucleoside (~ 3H-TdR) and ~ 3H-leucine incorporation were investigated to elucidate the relationship between free radicals and acidosis on VSMC proliferation.
