目的:优选山里黄根多糖的提取工艺。方法:设计单因素试验和正交试验,以多糖含量为指标,采用苯酚-硫酸法,比较影响多糖含量主要因素的主次关系,优化提取条件,确定最佳提取工艺。结果:影响山里黄根多糖提取率的主次因素为:料液比>提取次数>提取温度>提取时间;山里黄根多糖的最佳提取工艺条件为:提取温度100℃、提取时间100 min、料液比1∶10(g/m L)、提取次数3次,在此条件下,多糖提取率可达72.49 mg/g。结论:优选的山里黄根多糖提取工艺提取率较高,且方法稳定可行。
Objective: To optimize the extraction of polysaccharides from Radix Rehmanniae. Methods: Single factor experiment and orthogonal experiment were designed. The content of polysaccharide was used as index, phenol - sulfuric acid method was used to compare the main factors that affect the content of polysaccharide. The optimal extraction conditions were optimized and the optimum extraction process was determined. Results: The primary and secondary factors influencing the extraction rate of polysaccharides were as follows: solid-liquid ratio> extraction times> extraction temperature> extraction time; the optimum extraction conditions were as follows: extraction temperature 100 ℃, extraction time 100 min, The ratio of solid to liquid was 1:10 (g / m L), and the extraction time was 3 times. Under this condition, the extraction rate of polysaccharide reached 72.49 mg / g. Conclusion: The extraction rate of polysaccharides extracted from the root of Radix isatidis is high, and the method is stable and feasible.
