目的:研究紫草素对Ⅱ型胶原诱导的关节炎(CIA)MMP-1、TIMP-1表达的影响。方法:紫草素及对照药物口服给药CIA小鼠,连用30天,用酶联免疫吸附测定试剂盒测定CIA小鼠血清中的MMP-1、TIMP-1;用逆转录聚合酶链反应测定CIA小鼠髌骨及邻近滑膜组织的MMP-1、TIMP-1表达水平。结果:与模型组(775.9 102.12)相比,紫草素组(255.9 103.7)降低了CIA小鼠血清中MMP-1水平(P<0.001),紫草素组(135.3 9.1)与模型组(106.3 7.07)相比,升高了TIMP-1水平(P<0.05),髌骨及邻近滑膜组织的MMP-1mRNA水平与对照组相比显著被抑制(P<0.05);TIMP-1mRNA水平与对照组相比明显升高(P<0.05)。结论:紫草素可能通过抑制MMP-1,提高TIMP-1发挥对CIA小鼠的治疗作用。
Objective: To study the effect of shikonin on the expression of MMP-1 and TIMP-1 induced by type Ⅱ collagen in arthritis (CIA). Methods: CIA mice were administered orally with shikonin and control drugs for 30 days. Serum levels of MMP-1 and TIMP-1 in CIA mice were measured by enzyme linked immunosorbent assay (ELISA) kit. The levels of MMP-1 and TIMP-1 were determined by reverse transcription polymerase chain reaction The expression of MMP-1 and TIMP-1 in patella and adjacent synovial tissue of CIA mice. Results: Compared with model group (775.9 102.12), shikonin group (255.9 103.7) decreased serum MMP-1 level in CIA mice (P <0.001), shikonin group (135.3 9.1) and model group (P <0.05). The MMP-1 mRNA level in patella and adjacent synovial tissue was significantly inhibited compared with that in control group (P <0.05). The level of TIMP-1 mRNA in control group (P <0.05). Conclusion: Shikonin may increase the therapeutic effect of TIMP-1 on CIA mice by inhibiting MMP-1.
