目的探讨砷对周围血淋巴细胞中及人肺腺癌细胞系A549细胞中同源异型盒D10(HOXD10)基因表达的影响。方法 (1)采用判断抽样方法,抽取某砒霜厂职业性接触砷的59名工人为接触组,以无职业性接触砷的17名当地居民为对照组。采用氢化物发生-冷肼捕集-原子吸收法检测2组人群尿中形态砷水平,采用实时荧光定量聚合酶链式反应(qRT-PCR)法检测周围血淋巴细胞中HOXD10的表达。(2)以浓度为0.0、0.1、0.5、1.0、2.0μmol/L的三氧化二砷(As_2O_3)处理A549细胞,采用比色法检测细胞存活率,采用qRT-PCR法检测HOXD10的表达。结果 (1)接触组人群尿中无机砷、一甲基砷酸、二甲基砷酸、总砷水平和周围血淋巴细胞中HOXD10 mRNA相对表达水平均高于对照组(P<0.05)。(2)As_2O_3可导致A549细胞存活率呈现剂量依赖性下降(P<0.01),HOXD10mRNA相对表达水平呈现剂量依赖性上调(P<0.01)。A549细胞存活率和HOXD10 mRNA相对表达水平呈负相关,相关系数为-0.777(P<0.01)。结论砷可导致职业接触人群周围血中HOXD10表达上调。As_2O_3可抑制A549细胞增殖,可能与上调HOXD10表达有关。
Objective To investigate the effect of arsenic on the gene expression of homeobox D10 (HOXD10) in peripheral blood lymphocytes and human lung adenocarcinoma cell line A549. Methods (1) According to the sample sampling method, 59 workers who took arsenic in occupational exposure to arsenic in a cream factory were selected as the exposure group and 17 residents without occupational exposure to arsenic were used as the control group. The levels of urinary arsenic in the two groups were detected by hydride generation-cold hydrazine capture-atomic absorption spectrometry. The expression of HOXD10 in peripheral blood lymphocytes was detected by real-time quantitative polymerase chain reaction (qRT-PCR). (2) A549 cells were treated with different concentrations of As2O3 (0.0,0.1,0.5,1.0,2.0μmol / L), cell viability was detected by colorimetric assay, and the expression of HOXD10 was detected by qRT-PCR. Results (1) The levels of urinary inorganic arsenic, monomethyl arsenic acid, dimethyl arsenic acid, total arsenic and HOXD10 mRNA in the peripheral blood lymphocytes were higher than those in the control group (P <0.05). (2) As 2 O 3 could decrease the survival rate of A549 cells in a dose-dependent manner (P <0.01), and the relative expression level of HOXD10 mRNA increased in a dose-dependent manner (P <0.01). The survival rate of A549 cells was negatively correlated with the relative expression of HOXD10 mRNA, with a correlation coefficient of -0.777 (P <0.01). Conclusion Arsenic can cause HOXD10 expression in peripheral blood of occupationally exposed people. As_2O_3 can inhibit the proliferation of A549 cells, which may be related to the up-regulation of HOXD10 expression.
