目的:观察稳肾颗粒对慢性肾衰竭大鼠基质金属蛋白酶-9(MMP-9)、金属蛋白酶组织抑制因子-1(TIMP-1)和ERK通路表达的影响,初步探讨其改善肾功能的作用机制。方法:SD大鼠21只,随机分为正常组、模型组、治疗组。采取腺嘌呤灌胃造模,予稳肾颗粒干预2周后处死,留取标本。Real time-PCR测MMP-9及TIMP-1含量;WB测ERK、p-ERK蛋白含量。结果:稳肾颗粒可以改善模型鼠肾功能、上调MMP-9,下调TIMP-1、p-ERK。结论:稳肾颗粒可能通过抑制ERK信号通路、调节MMP-9及TIMP-1表达来改善肾功能。
Objective: To observe the effect of “Wen Shen Granule” on the expression of matrix metalloproteinase-9 (MMP-9), tissue inhibitor of metalloproteinase-1 (TIMP-1) and ERK pathway in rats with chronic renal failure mechanism. Methods: Twenty-one SD rats were randomly divided into normal group, model group and treatment group. Take adenine gavage modeling, to stabilize the kidney particles were sacrificed 2 weeks after the intervention, specimens were taken. The contents of MMP-9 and TIMP-1 were detected by Real time-PCR. The contents of ERK and p-ERK were measured by WB. Results: SF can improve renal function, up-regulate MMP-9, down-regulate TIMP-1 and p-ERK. Conclusion: SF can improve renal function by inhibiting the ERK signaling pathway and regulating the expression of MMP-9 and TIMP-1.
