目的观察中药祛斑霜对雄性棕黄色豚鼠皮肤中超氧化物岐化酶(SOD)、丙二醛(MDA)和黑素颗粒的影响。方法将40只雄性棕黄色豚鼠随机分为空白组、模型组、阳性对照组、中药祛斑霜组4组,每组10只。在各组豚鼠背部取3 cm×3 cm皮肤进行隔日脱毛1次。除空白组以外,其他各组以波长为320 nm的中波紫外线每日照射1次,每次照射时间为60 min,动物与光源的距离为10 cm,连续照射4周,连续观察各组豚鼠背部脱毛部位皮肤变化情况。造模成功后,中药祛斑霜组、阳性对照组分别予相应药物外涂于各豚鼠皮损处,每日1次,每次按摩5 min。连续给药4周后,动物断颈处死,迅速取其脱毛后皮肤组织。检测皮肤中SOD活性和MDA值,通过HE染色对黑素颗粒进行镜下观察;采用免疫组化法标记黑素颗粒并测定其阳性表达区域的平均光密度,分析黑素颗粒的阳性表达水平。结果中药祛斑霜组SOD明显高于模型组和阳性对照组(P<0.05),而MDA明显低于模型组和阳性对照组(P<0.05),并且中药祛斑霜组黑色颗粒表达明显低于模型组和阳性对照组(P<0.05)。结论中药祛斑霜可以提高黄褐斑豚鼠模型皮肤中SOD的活性,降低MDA的含量和减少皮肤中黑素颗粒的生成。
Objective To observe the effect of freckle cream on superoxide dismutase (SOD), malondialdehyde (MDA) and melanin granules in the skin of male brown-yellow guinea pigs. Methods Forty male, yellow-brown guinea pigs were randomly divided into 4 groups: blank group, model group, positive control group and traditional Chinese medicine freckle cream group, with 10 rats in each group. In the back of each group of guinea pigs to take 3 cm × 3 cm skin hair removal every other day. Except for the blank group, the other groups were irradiated with UVB at a wavelength of 320 nm once a day for 60 min, distance between the animals and the light source was 10 cm, and continuous irradiation for 4 weeks. The guinea pigs Back hair removal site skin changes. After the success of modeling, Chinese medicine freckle cream group, the positive control group were given the corresponding drug applied to the guinea pig lesions, 1 day, each massage 5 min. After 4 consecutive weeks of administration, the animals were sacrificed and their epidermis removed immediately after epilation. The activity of SOD and the MDA value in the skin were detected. The melanin granules were observed under the microscope by HE staining. The melanin granules were detected by immunohistochemistry and the average optical density was measured. The positive expression of melanin granules was analyzed. Results The SOD in the Chinese medicine freckle cream group was significantly higher than that in the model group and the positive control group (P <0.05), while the MDA content in the freckle cream group was significantly lower than that in the model group and the positive control group (P <0.05) Group and positive control group (P <0.05). Conclusion Chinese medicine freckle cream can increase the activity of SOD in cutaneous guinea pig model, reduce the content of MDA and reduce the production of melanin granules in the skin.
