目的:探讨舌鳞癌(TSCC)中细胞质膜微囊蛋白-1(caveolin-1)基因及蛋白的表达水平及其与肿瘤生物学特性的关系。方法:应用原位杂交和免疫组化法,测定92例TSCC患者舌体癌组织及癌周正常舌组织中caveolin-1基因及其蛋白表达水平变化,应用SPSS10.0软件包对数据进行统计学分析。结果:TSCC中caveolin-1基因mRNA表达水平(24.6±1.8)及蛋白水平(471.2±68.1)分别低于正常组织的167.2±20.5及964.5±77.2,差异均有统计学意义(P<0.01)。高分化TSCC中,caveolin-1蛋白表达水平为5.6±1.4,高于中、低分化的1.3±0.8,差异有统计学意义(P<0.01);临床Ⅰ~Ⅱ期表达水平为6.1±2.0,高于临床Ⅲ~Ⅳ期的2.4±0.7,差异有统计学意义(P<0.01)。颈淋巴结转移与caveolin-1蛋白表达水平无关(P>0.05)。结论:TSCC组织中caveolin-1基因及蛋白的表达明显下调,caveolin-1蛋白的表达与肿瘤的分化程度、临床分期有关,而与肿瘤转移无关。
Objective: To investigate the expression of caveolin-1 gene and protein in tongue squamous cell carcinoma (TSCC) and its relationship with tumor biological characteristics. Methods: The in situ hybridization and immunohistochemical methods were used to detect the changes of caveolin-1 gene and its protein expression in tongue cancer tissues and normal tongue tissues in 92 TSCC patients. The data were analyzed by SPSS 10.0 software package analysis. Results: The caveolin-1 mRNA expression level (24.6 ± 1.8) and protein level (471.2 ± 68.1) in TSCC were significantly lower than those in normal tissues (167.2 ± 20.5 and 964.5 ± 77.2, respectively) (P <0.01). The expression of caveolin-1 in well-differentiated TSCC was 5.6 ± 1.4, which was significantly higher than that in moderate and poorly differentiated 1.3 ± 0.8 (P <0.01). The expression of caveolin-1 in stage Ⅰ-Ⅱ was 6.1 ± 2.0, Higher than that of clinical stage Ⅲ ~ Ⅳ 2.4 ± 0.7, the difference was statistically significant (P <0.01). The cervical lymph node metastasis had no correlation with caveolin-1 protein expression (P> 0.05). Conclusion: The expression of caveolin-1 gene and protein in TSCC was significantly down-regulated. The expression of caveolin-1 was related to the differentiation degree and clinical stage of TSCC, but not to tumor metastasis.
