为探讨中药水银(Hg)和轻粉(HgCl)的抗肿瘤作用,以三氧化二砷(AS2O3)及阿糖胞苷(Ara-c)为对照,应用细胞形态学、流式细胞术、MTT法和药物反应曲线等方法观察3种药物对K562和HEL细胞株的作用。结果表明,HgCl2和As2O3均能诱导562细胞凋亡,而Ara-c无此作用.HgCl2可使K(562)的G0/G1增高,提示激活P53基因可能是氯化汞诱导凋亡机制之一;HgCl2和AS2O3均可显著抑制K(562)增殖;Ara-c抑制HEL增殖作用明显强于HgCl2和As2O3三者对HEL均无诱导凋亡作用,但对K(562)和HEL均有不同程度杀伤和致死作用。HgCl2和As2O3作用相似,其主要机制可能是与Hg(2+)和AS(3+)与巯基(-SH)有极高亲和力,继而与含巯基的活性基结合、降低诸多酶的活性、损害细胞膜有关。HgCl2具有抗肿瘤作用,是一种新的诱导细胞凋亡药物。
To investigate the anti-tumor effects of Chinese medicine mercury (Hg) and light powder (HgCl), we used AS2O3 and Ara-c as controls to apply cell morphology, flow cytometry, MTT assay, and drugs. The response curves and other methods were used to observe the effects of the three drugs on K562 and HEL cell lines. The results showed that both HgCl2 and As2O3 could induce apoptosis of 562 cells, while Ara-c had no such effect. HgCl2 increased the G0/G1 of K(562), suggesting that activating P53 gene may be one of the mercury chloride-induced apoptosis mechanisms. Both HgCl2 and AS2O3 could significantly inhibit the proliferation of K(562); Ara-c inhibited the proliferation of HEL significantly more than HgCl2 and As2O3, but did not induce apoptosis in HEL, but had different degrees of K(562) and HEL.
