人参总皂苷对慢性心力衰竭大鼠血流动力学及caspase-3表达的影响

目的研究人参总皂苷(GS)对慢性心力衰竭大鼠血流动力学、血浆脑钠肽(BNP)、心肌细胞内钙离子浓度的影响及其作用机制。方法雄性清洁级Wistar大鼠50只,分为对照组、模型组、人参总皂苷(GS)3个剂量组。对照组和模型组每日灌胃蒸馏水40 m L;3个剂量GS组分别灌胃人参总皂苷5,20,80mg·kg~(-1),qd,连续4周。测定各组大鼠血流动力学变化,血浆脑钠肽(BNP)及心肌细胞内Ca~(2+)浓度。结果与对照组比较,模型组和GS组,左心室收缩压(LVSP)、左心室内压最大下降速率(-dp/dtmax)显著降低(P<0.05),且GS组显著高于模型组(P<0.05);与对照组比较,模型组和GS组的左心室舒张末期压(LVEDP)、左心室内压最大上升速率(+dp/dtmax)显著升高(P<0.05),GS组显著低于模型组(P<0.05);与对照组比较,模型组和GS组血浆BNP、心肌细胞内Ca~(2+)均明显升高(P<0.05),GS组显著低于模型组(P<0.05);与模型组比较,对照组和GS组的大鼠心肌细胞Caspase-3蛋白表达明显降低(P<0.05),但GS组显著高于对照组(P<0.05)。结论人参总皂苷可改善心力衰竭大鼠血流动力学指标,降低心肌细胞钙离子浓度。其作用机制可能与抑制心肌细胞Caspase-3表达有关。 Objective To study the effects of ginseng total saponin (GS) on hemodynamics, plasma brain natriuretic peptide (BNP) and intracellular calcium concentration in rats with chronic heart failure and its mechanism. Methods Fifty male Wistar rats were divided into three groups: control group, model group and total ginsenoside (GS). Control group and model group were fed with distilled water 40 m L daily; GS 3 groups were gavaged with gypenosides 5, 20 and 80 mg · kg -1, qd for 4 weeks respectively. The changes of hemodynamics, plasma brain natriuretic peptide (BNP) and intracellular Ca2 + concentration in myocardial cells were determined. Results Compared with the control group, the left ventricular systolic pressure (LVSP) and the maximal rate of left ventricular pressure (-dp / dtmax) decreased significantly in model group and GS group (P <0.05), and GS group was significantly higher than that in model group (P <0.05). Compared with the control group, the left ventricular end-diastolic pressure (LVEDP) and the maximum rate of left ventricular pressure (+ dp / dtmax) in model group and GS group were significantly increased (P <0.05) .Compared with the control group, plasma BNP and Ca 2+ in myocardial cells in model group and GS group were significantly increased (P <0.05), while those in GS group were significantly lower than those in model group P <0.05). Compared with the model group, Caspase-3 protein expression in the control group and GS group was significantly decreased (P <0.05), but GS group was significantly higher than that in the control group (P <0.05). Conclusion Ginseng total saponins can improve hemodynamic parameters of heart failure rats, reduce the concentration of calcium in myocardial cells. Its mechanism may be related to the inhibition of myocardial cell Caspase-3 expression.
