从日本血吸虫感染兔红细胞上分离的SjR47和SjR12两种组分蛋白能诱导宿生产生明显的抗病免疫效应。为大量纯化、收集该两种组分蛋白,我们采用了聚丙烯酰胺凝胶大柱制备电泳技术,并对该法收集蛋白的纯度及其生物活性进行了鉴定。结果表明,该技术不仅可使蛋白收集量达到毫克级水平,而且蛋白纯度较高,还可保持较高的天然蛋白生物活性,经透析处理后蛋白生物活性更高,用以免疫家兔后可获得高效价的多克隆抗体,为今后进一步深入研究该两种蛋白分子的特性奠定了基础。实验结果进一步表明聚丙烯酰胺凝胶大柱制备电泳技术是一种有效的蛋白纯化方法。
SjR47 and SjR12 proteins isolated from rabbit erythrocytes infected with Schistosoma japonicum could induce significant anti-disease and immunogenic effects on the host.
