目的:探讨肢体缺血再灌注(limbs ischemia-reperfusion,LIR)对脑组织的影响。方法:复制大鼠LIR模型,采用尼氏染色观察脑组织病理学改变,TUNEL法检测细胞凋亡情况,免疫组化和Western印迹法检测凋亡相关因子Bcl-2、细胞色素C(cyto C)、Caspase-3的表达变化。结果:大鼠LIR后,脑组织海马区、中脑红核区神经元细胞受损,脑组织出现水肿裂隙,cyto C、 Caspase-3凋亡相关蛋白表达明显上调,24 h达高峰.与细胞凋亡数量的增加相一致,Bcl-2表达与再灌时程呈负相关。结论:LIR 后可导致脑组织损伤,并出现细胞凋亡,其发生机制与Caspase-3、cyto C、Bcl-2凋亡相关蛋白的表达变化有关。
Objective: To investigate the effect of limbs ischemia-reperfusion (LIR) on brain tissue. Methods: The rat model of LIR was duplicated, the pathological changes of brain were observed by Nissl staining, the apoptosis was detected by TUNEL, the expression of apoptosis related factors Bcl-2, cyto C, , Caspase-3 expression changes. Results: After LIR in rats, the neuronal cells in the hippocampus and midbrain red nucleus were impaired, and the edema and fissure in the brain tissue were observed. The expressions of cyto C, Caspase-3 apoptosis-related proteins were significantly up-regulated and peaked at 24 h. Consistent with the increase in the number of apoptotic cells, the expression of Bcl-2 was negatively correlated with the course of reperfusion. CONCLUSION: LIR can induce brain injury and induce apoptosis. The mechanism may be related to the changes of the expression of apoptosis-related proteins Caspase-3, cyto C and Bcl-2.
