目的:观察消膏转浊方对高脂饲料喂养的肥胖大鼠肝脏肿瘤坏死因子-α(TNF-α)和瘦素(Leptin)与胰岛素抵抗的相关性。方法:SPF级Wistar雄性大鼠40只随机分为5组各8只,正常组以普通饲料喂养,其余组的大鼠以高脂饲料喂养。8周后,正常组仍以普通饲料喂养;模型组继续以高脂饲料喂养;消膏转浊组以及罗格列酮组改为普通饲料喂养,同时给予药物治疗,饮食控制组改为普通饲料喂养。共为12周。于禁食次日进行钳夹实验并计算葡萄糖输注率(GIR);检测大鼠肝脏胰岛素受体(InsR)和胰岛素受体底物-2(IRS-2)的基因表达;检测肝匀浆中TNF-α、Leptin的含量。结果:与模型组比较,消膏转浊组TNF-α和Leptin的含量降低(P<0.05,P<0.01);消膏转浊组与罗格列酮组GIR显著提高(P<0.01);消膏转浊组与罗格列酮组InsR mRNA、IRS-2 mRNA的表达均显著高于模型组(P<0.01)。结论:消膏转浊方可以逆转肝脏细胞脂肪变性,其机制可能是通过降低肝脏TNF-α的过度表达,改善Leptin抵抗,来达到进一步减少肝脏胰岛素抵抗。
Objective: To observe the effect of Xiaoganzhuanzhuo prescription on TNF-α, Leptin and insulin resistance in obese rats fed with high fat diet. Methods: Forty SPF Wistar male rats were randomly divided into 5 groups, 8 rats in each group. Normal rats were fed with normal diet, and the rest rats were fed with high fat diet. After 8 weeks, the normal group was still fed with normal diet; the model group continued to be fed with high-fat diet; the group with Xiaogan Zhuanzhuo and the rosiglitazone group was changed to ordinary feed while the drug treatment was given. The diet control group was changed to common feed feed. A total of 12 weeks. The rats were subjected to the clamping test on the next day after fasting and the glucose infusion rate (GIR) was calculated. The gene expression of rat liver insulin receptor (InsR) and insulin receptor substrate-2 (IRS-2) In the TNF-α, Leptin content. Results: Compared with the model group, the levels of TNF - α and Leptin in Xiaogan Zhuanzhuo group decreased significantly (P <0.05, P <0.01); the GIR of Xiaoguazhuo turbid group and rosiglitazone group increased significantly (P <0.01); The expressions of InsR mRNA and IRS-2 mRNA in Xiaogan Zhuanzhuo group and rosiglitazone group were significantly higher than those in model group (P <0.01). Conclusion: Xiaogan Zhuanzhuo can reverse the fatty degeneration of liver cells, and its mechanism may be to further reduce the hepatic insulin resistance by reducing the over expression of TNF-α and improving the Leptin resistance.
