目的研究血必净注射液对急性肝损伤大鼠肝组织中过氧化物酶体增殖物激活受体γ(PPAR-γ)表达的动态影响。方法将SD大鼠随机分为对照组、模型组和治疗组,采用腹腔注射D-氨基半乳糖(D-Gal)、脂多糖建立急性肝损伤大鼠模型。治疗组在造模前6 h及制模后大鼠腹腔内给予血必净注射液2.5 g/kg,12 h 1次,共4次,各组于造模后6、12、24、48 h 4个时间点留取大鼠血及肝脏标本。观察大鼠肝功能及肝脏病理变化;用RT-PCR法测定各组肝PPAR-γmRNA表达水平;ELISA法测定血清中肿瘤坏死因子-α(TNF-α),白细胞介素-6(IL-6)水平。结果模型组大鼠肝组织PPAR-γmRNA表达明显低于对照组,治疗组组织大鼠肝PPAR-γmRNA水平明显高于模型组,组间比较差异有统计学意义。结论血必净注射液对实验性急性肝损伤有一定保护作用,其机制可能与升高鼠肝PPAR-γmRNA表达水平有关。
Objective To study the dynamic effects of Xuebijing injection on the expression of peroxisome proliferator - activated receptor γ (PPAR - γ) in liver of acute liver injury rats. Methods SD rats were randomly divided into control group, model group and treatment group. The model of acute liver injury was established by intraperitoneal injection of D-Gal and lipopolysaccharide. In the treatment group, Xuebijing Injection 2.5 g / kg and 12 h were given intraperitoneally once a day for 6 h before model establishment and 4 times in all groups. After 6, 12, 24 and 48 h Four time points to take rat blood and liver specimens. The liver function and pathological changes of liver were observed. The level of PPAR-γmRNA in liver of model group was significantly higher than that of model group, the difference was statistically significant. Conclusion Xuebijing Injection may have a protective effect on experimental acute liver injury, and its mechanism may be related to increasing the expression of PPAR-γmRNA in rat liver.
