建立了同时测定大鼠胆汁中 4种盐酸非洛普 ( DDPH) 相代谢物的 HPLC方法。胆汁样品经盐酸水解后以 Alltech C1 8固相萃取小柱纯化富集 ,然后进行 HPLC分析。色谱柱为 Hypersil ODS2 1 0μ 30 cm× 4 .6mm ID,流动相为甲醇 -水 -三乙胺 -冰乙酸 ( 450∶ 550∶ 1∶ 0 .7) ,检测波长为 2 78nm。测定了 ip DDPH后大鼠胆汁中 4个 DDPH代谢物的体内动态。结果表明 M1和 M2为 DDPH主要体内代谢产物。给药后 5h DDPH代谢产物的胆汁累积排泄量已达胆汁总排泄量的 75%~ 80 %。给药后 0 .5~ 1 h时间段 DDPH代谢物的胆汁排泄速率达峰值
A HPLC method was established for the simultaneous determination of four metabolites of felodipine hydrochloride (DDPH) in rat bile. The bile samples were hydrolyzed with hydrochloric acid and purified on an Alltech C1 8 solid phase extraction cartridge, followed by HPLC analysis. The chromatographic column was Hypersil ODS2 1 0μ 30 cm × 4.6mm ID with a mobile phase of methanol-water-triethylamine-glacial acetic acid (450: 550: 1: 0.7) at a detection wavelength of 2 78nm. The in vivo dynamics of 4 DDPH metabolites in rat bile after ip DDPH were measured. The results show that M1 and M2 are the major metabolites of DDPH in vivo. 5h after administration of DDPH metabolites cumulative excretion of bile has reached 75% to 80% of the total bile excretion. DDPH metabolites reached the peak bile excretion rate at 0.5-1 h after administration
