目的 了解转化生长因子β1(TGF- β1)及其受体(TβR)在绒毛及蜕膜组织中的表达,初步探讨TGF- β1与体外受精胚胎移植(IVF- ET)后流产及自然流产的关系。方法 应用免疫组化链霉菌抗生物素蛋白过氧化物酶(SP)连接法,检测10例IVF -ET后流产患者(IVF- ET组)和20例早期自然流产患者(自然流产组)的绒毛及蜕膜组织中的TGF -β1及TβR(Ⅰ、Ⅱ型)表达情况,其表达水平以阳性染色部位的吸光度(A)值表示,并与20例人工流产者(对照组)进行比较。结果 TGF- β1及TβR在早孕绒毛滋养细胞、蜕膜组织腺体及间质细胞中均有阳性染色, 3组表达部位相同。TGF β1在IVF ET组蜕膜及绒毛组织中表达水平(A值,中位数,下同)分别为0 167、0 .199,自然流产组为0 .198、0 .201,对照组为0 .277、0 .274;TβR Ⅰ在IVF- ET组蜕膜及绒毛组织中表达水平为0 144、0 150,自然流产组为0 .202、0. 201,对照组为0 .238、0 .281;TβR Ⅱ在IVF ET组蜕膜及绒毛组织中的表达水平为0 .199、0 .145,自然流产组为0. 153、0 .156,对照组为0 .300、0 .301。上述指标,IVF -ET、自然流产组分别与对照组比较,差异均有统计学意义(P<0. 01)。结论 TGF -β1及TβR在IVF- ET后流产及自然流产患者绒毛及蜕膜组织中的表达下降,在IVF- ET后流产发病中起重要作用,?
Objective To investigate the expression of transforming growth factor β1 (TGF-β1) and its receptor (TβR) in villi and decidua and to explore the relationship between TGF-β1 and abortion and spontaneous abortion after IVF-ET . Methods Using the immunohistochemical Streptavidin-Peroxidase (SP) ligation method, the villi of 10 cases of abortion after IVF-ET (IVF-ET group) and 20 cases of spontaneous abortion (spontaneous abortion group) And decidual tissue of TGF-β1 and TβR (Ⅰ, Ⅱ type) expression, the expression level of positive staining of the absorbance (A) values, and with 20 cases of induced abortion (control group) were compared. Results TGF-β1 and TβR in the first trimester villous trophoblast cells, decidual glandular cells and interstitial cells were positive staining, the three groups expressed the same site. The expression level of TGFβ1 in the decidua and villus of IVF ET group (A value, median, the same below) were 0 167,0199, spontaneous abortion group was 0.198,0.201, the control group was 0 .277,0. 274; The expression level of TβR Ⅰ in the decidua and villus tissues of IVF-ET group was 0 144,0 150, that of spontaneous abortion group was 0.202,0.201 and that of the control group was 0.238,0. 281; The expression level of TβR Ⅱ in the decidua and villus tissues of IVF ET group was 0.199,0. 145, that of spontaneous abortion group was 0.153, 0.156 and that of the control group was 0.300, 0.301. The above indicators, IVF-ET, spontaneous abortion group compared with the control group, the difference was statistically significant (P <0.01). Conclusion The decreased expression of TGF-β1 and TβR in the villi and decidua of patients with abortion and spontaneous abortion after IVF-ET play an important role in the abortion after IVF-ET.
