目的:建立HPLC法同时定量分析酸橙果实类药材中8个黄酮类成分(芸香柚皮苷、柚皮苷、橙皮苷、新橙皮苷、柚皮素、橙皮素、川陈皮素和桔皮素)和3个香豆素类成分(水合橙皮内酯、马尔敏及6’,7’-二羟基香柠檬亭)。方法:色谱柱为Phenomenex C18(250 mm×4.6 mm,5μm);流动相为乙腈-水,梯度洗脱;流速1.0 m L/min;检测波长320 nm;柱温30℃。结果:8个黄酮类成分与3个香豆素类成分分别在6.185~247.4μg/m L、53.4~2 136.9μg/m L、2.525~101.0μg/m L、26.153~1 046.1μg/m L、0.9765~38.7μg/m L、0.9825~39.3μg/m L、1.56~62.4μg/m L、1.485~59.4μg/m L、3.17~126.8μg/m L、1.035~41.4μg/m L、0.815~32.6μg/m L范围内与峰面积具有良好的线性关系(r≥0.9996);平均加样回收率(n=6)分别为104.74%、103.92%、97.46%、98.76%、103.94%、97.68%、99.74%、104.35%、104.23%、101.60%、103.26%,RSD均小于2.5%(n=6)。结论:该含量测定方法可为酸橙及其近缘植物果实类药材的质量控制提供参考。
OBJECTIVE: To establish a HPLC method for the simultaneous determination of eight flavonoids in fruits of lime fruits (ruthenium, naringin, hesperidin, neohesperidin, naringenin, hesperetin, nobiletin and Orange peel) and three coumarins (hydrated orange lactone, margarine and 6 ’, 7’-dihydroxybenzanthaminer). Methods: The column was Phenomenex C18 (250 mm × 4.6 mm, 5 μm). The mobile phase was acetonitrile-water gradient elution. The flow rate was 1.0 m L / min. The detection wavelength was 320 nm and the column temperature was 30 ℃. Results: The contents of 8 flavonoids and 3 coumarins were 6.185 ~ 247.4μg / m L, 53.4 ~ 2 136.9μg / m L, 2.525 ~ 101.0μg / m L, 26.153 ~ 1 046.1μg / m L , 0.9765 to 38.7 μg / m L, 0.9825 to 39.3 μg / m L, 1.56 to 62.4 μg / m L, 1.485 to 59.4 μg / m L, 3.17 to 126.8 μg / m L, 1.035 to 41.4 μg / m L, 0.815 (R≥0.9996). The average recoveries (n = 6) were 104.74%, 103.92%, 97.46%, 98.76%, 103.94%, 97.68 %, 99.74%, 104.35%, 104.23%, 101.60%, 103.26%, respectively. All the RSDs were less than 2.5% (n = 6). Conclusion: This method can provide reference for the quality control of fruits and vegetables of lime and its related plants.
