目的研究化瘀理肺方对大鼠肺纤维化形成的干预作用及可能作用机制。方法将大鼠随机分为对照组、模型组、中药组、激素组,用1%戊巴比妥钠(35mg/kg)腹腔内注射麻醉后,经气管内插管注入博莱霉素(5mg/kg)建立肺纤维化模型,对照组注入生理盐水0.25ml,从造模后的第2天起分别给予化瘀理肺方药物灌胃(中药组,13410mg/kg)、生理盐水灌胃(对照组、模型组)、氢化可的松琥珀酸钠腹腔注射(激素组,25mg/kg)进行干预,后于给药后的第14、28天分别处死,取肺组织行苏木素-伊红(HE)染色观察病理变化情况,免疫组化法测定肺组织中Smad7蛋白的表达情况。结果中药组肺泡炎和纤维化程度明显低于模型组和激素组,中药组Smad7蛋白表达明显高于模型组和激素组(P<0.01)。结论化瘀理肺方对博莱霉素所致肺纤维化大鼠有一定预防性治疗作用,可能通过促进Smad7蛋白表达发挥作用。
Objective To study the intervention effect of Huayu Lifang Recipe on pulmonary fibrosis in rats and its possible mechanism. Methods Rats were randomly divided into control group, model group, traditional Chinese medicine group and hormone group. After intraperitoneal injection of sodium pentobarbital (1%, 35 mg / kg), anesthesia was induced by intraperitoneal injection of bleomycin / kg) to establish the model of pulmonary fibrosis, the control group was injected with saline 0.25ml, respectively, from the second day after modeling were given Huayu Lifeang Fang gavage (Chinese medicine group, 13410mg / kg), saline (Control group, model group), intraperitoneal injection of hydrocortisone sodium succinate (hormone group, 25mg / kg) for intervention, and then sacrificed on the 14th and 28th day after administration respectively. The lungs were subjected to hematoxylin-eosin HE staining and pathological changes were observed. The expression of Smad7 in lung tissue was detected by immunohistochemistry. Results The degree of alveolitis and fibrosis in the TCM group was significantly lower than that in the model group and the hormone group. The Smad7 protein expression in the TCM group was significantly higher than that in the model group and the hormone group (P <0.01). Conclusion Huayu Lifang Recipe has certain preventive effect on bleomycin-induced pulmonary fibrosis in rats and may play a role in promoting Smad7 protein expression.
