目的 建立HLA新基因B 5 6 10携带者家系的B淋巴细胞系 ,用于该基因的研究和确证。方法 采集外周血分离淋巴细胞 ,用EBV感染淋巴细胞后 ,加入含 2 0 %FBS、2 μg/mlCsA的RPMI16 4 0进行培养。 结果 5位家庭成员的无限增殖化B淋巴细胞系新基因稳定遗传。结论 建立的无限增殖化B淋巴细胞系的HLA新基因B 5 6 10遗传稳定 ,为大量保存和繁殖珍贵的生物医学材料奠定了基础。
OBJECTIVE: To establish a B lymphocyte lineage of the HLA-B 5 6 10 pedigree family for the study and confirmation of this gene. Methods Lymphocytes were isolated from peripheral blood and infected with EBV. The cells were cultured with RPMI1640 containing 20% FBS and 2 μg / ml CsA. Results The five members of the immortalized B lymphocyte cell line were genetically stable. Conclusion The established genetic immortalized B 5 6 10 immortalized B lymphocyte cell line lays the foundation for the preservation and multiplication of valuable biomedical materials.
