为探讨滇杨分枝特性差异的分子机理,对采集于四川省和云南省的59个滇杨优树无性系3a生苗木进行了cDNA-AFLP差异研究,分析了苗木侧芽休眠时基因表达的多样性。结果表明:筛选出的7对引物组合共扩增出了总条带608条,其中有353条差异性条带,Nei’s基因多样性指数为0.1061,Shannon信息指数为0.1856,遗传分化系数为0.0123,居群间的基因流为40.28,表明居群间的遗传分化程度较低。分子生物学方差分析(AMOVA)结果表明,居群间的差异仅为0.99%,差异不显著(P=0.052),居群内差异占99.01%,差异极显著;表明滇杨侧芽在休眠期个体之间基因表达的复杂性;聚类分析结果显示,在亚分枝单元或最小分枝单元,大部分无性系能够以分枝性状相聚。该研究结果为进一步开展滇杨分枝相关基因的筛选及克隆等工作奠定了前期研究基础。
In order to explore the molecular mechanism of the difference in branching characteristics of Populus davidiana, the differences of cDNA-AFLP among 59 Populus euphratica clones collected in Sichuan and Yunnan Provinces were studied. The diversity of gene expression during the dormancy of shoots was analyzed. The results showed that there were 608 total bands amplified by 7 pairs of primers, including 353 differential bands, Nei’s gene diversity index of 0.1061, Shannon’s information index of 0.1856, genetic differentiation coefficient of 0.0123, Gene flow between populations was 40.28, indicating a low degree of genetic differentiation among populations. The results of molecular analysis of variance (AMOVA) showed that the difference between populations was only 0.99%, with no significant difference (P = 0.052), and the difference among populations was 99.01% The results of cluster analysis showed that in the sub-branching units or the smallest branching units, most of the clones can be clustered with branching traits. The results of this study laid the foundation of previous research for further screening and cloning related genes of D. yunnanensis.
