为探讨血管平滑肌细胞自分泌和旁分泌对血管平滑肌细胞增殖的抑制作用 ,采用家兔主动脉贴块培养法培养血管平滑肌细胞 ,并制备平滑肌细胞条件培养基。采用孔径 10 0kDa和 10kDa的Millipore滤膜 ,将平滑肌细胞条件培养基分部。平滑肌细胞增殖测定采用宝灵曼试剂盒XTT法。结果提示 ,家兔血管平滑肌细胞条件培养基抑制血管平滑肌细胞的增殖 ,浓度 5 0 %的平滑肌细胞条件培养基抑制血管平滑肌细胞的增殖率高达 45 % ,且该生长抑制活性物质具有剂量依赖、加热 5 6℃ 30min稳定和分子质量小于 10kDa的特点。
In order to investigate the inhibitory effect of autocrine and paracrine on the proliferation of vascular smooth muscle cells, vascular smooth muscle cells (VSMCs) were cultured in rabbit aorta patch culture medium and conditioned medium of smooth muscle cells was prepared. Smooth muscle cell conditioned medium fractions were fractionated using Millipore filters with pore sizes of 100 kDa and 10 kDa. Smooth muscle cell proliferation assay using Bauman Kit XTT method. The results suggest that the conditioned medium of rabbit vascular smooth muscle cells inhibits the proliferation of vascular smooth muscle cells. The conditioned medium of smooth muscle cells in a concentration of 50% inhibits the proliferation rate of vascular smooth muscle cells up to 45%, and the growth inhibitory active substance has a dose-dependent, 5 6 ℃ 30min stable and molecular weight less than 10kDa characteristics.
