根据鲉形目鱼类Cytb和COⅡ上下游保守序列各自设计引物,对日本鬼鲉的线粒体DNA进行PCR扩增,克隆并测定了Cytb和COⅡ因的全序列,COⅡ基因为691bp,Cytb为1141bp。从GenBank下载28种鲉形目及外群鱼类的Cytb部分或全序列和10种鲉形目及外群鱼类的COⅡ部分或全序列,然后进行比对分析,用NJ法和ML法构建分子系统发育树。系统发育树结果显示:线粒体Cytb和COⅡ基因序列构建的分子系统树,与传统分类方法的结果基本一致;毒鲉科的日本鬼与科的关系最近;在飞角鱼科(Dactylopteridae)是否属于形目还是单独成豹鲂鲉目的问题上,本研究倾向支持飞角鱼科独立归成豹鲂鲉目(Dactylopteriformes)。日本鬼鲉的COⅡ与昆虫类的不同,形目的COⅡ基因比Cytb基因保守,即COⅡ基因比Cytb基因进化慢。COⅡ基因进化树相应的节点上数值稍高。
The mitochondrial DNA was amplified by PCR from Cytb and CO Ⅱ upstream and downstream conserved sequences. The complete sequence of Cytb and COⅡ gene was cloned and sequenced. The COⅡ gene was 691 bp and the Cytb was 1141 bp. The partial or complete sequence of Cytb and the partial sequence of Cytb from 28 species and their out-fishes were downloaded from GenBank and the COⅡ partial sequences or complete sequences of 10 species and their relatives were analyzed and compared by NJ and ML methods Molecular phylogenetic tree. The results of phylogenetic tree showed that the molecular phylogenetic trees of Cytb and COⅡ gene sequences of mitochondria were basically the same as those of the traditional taxonomic method. The relationship between Dodecylidae and Daphnida was the closest in Japan. On the other hand, this study tended to support that the Acinonyx family independently belong to Dactylopteriformes. The COⅡ gene of Japanese ghost beetles is different from that of insects. The shape of COⅡ gene is more conservative than that of Cytb gene, that is, the COⅡ gene is slower than the Cytb gene. The corresponding node of the evolutionary tree of COⅡ gene is slightly higher.
