目的:以胶原诱导性关节炎(CIA)大鼠为模型,观察洛伐他汀对CIA大鼠血清细胞因子的影响,为探索炎性关节炎新的治疗药物提供理论基础。方法:利用牛Ⅱ型胶原建立Wistar大鼠CIA模型,造模成功后随机分为空白对照组、CIA模型组、布洛芬组、甲氨蝶呤(MTX)组和洛伐他汀组。分别在第2周和第5周时用ELISA方法检测各组大鼠血清的肿瘤坏死因子α(TNF-α)、白介素6(IL-6)、白介素8(IL-8)和干扰素诱生蛋白-10(IP-10)的水平;用实时荧光定量PCR方法检测第5周时大鼠白介素1β(IL-1β)和白介素17(IL-17)mRNA的表达量。结果:免疫后第5周,洛伐他汀组、布洛芬组和MTX组大鼠的血清TNF-α,IL-6和IP-10的水平显著低于CIA组,而洛伐他汀组和MTX组的IL-8水平与CIA组大鼠无显著差异。洛伐他汀组和MTX组的IL-1β和IL-17 mRNA的表达量显著低于CIA组。结论:洛伐他汀能抑制TNF-α,IL-6,IL-1β,IL-17和IP-10的水平。
OBJECTIVE: To observe the effect of lovastatin on serum cytokines in CIA rats by using collagen induced arthritis (CIA) rats as a model to provide a theoretical basis for exploring new therapeutic agents for inflammatory arthritis. Methods: The CIA model of Wistar rats was established with bovine type Ⅱ collagen. The rats were randomly divided into blank control group, CIA model group, ibuprofen group, methotrexate (MTX) group and lovastatin group. Serum levels of TNF-α, IL-6, IL-8 and IFN-γ were detected by enzyme linked immunosorbent assay (ELISA) at week 2 and week 5 respectively The expression of interleukin-1β (IL-1β) and interleukin-17 (IL-17) mRNA in the fifth week was detected by real-time fluorescence quantitative PCR. Results: The levels of TNF-α, IL-6 and IP-10 in lovastatin group, ibuprofen group and MTX group were significantly lower than those in CIA group at the 5th week after immunization, while lovastatin group and MTX There was no significant difference between the group of IL-8 and CIA rats. The expressions of IL-1β and IL-17 mRNA in lovastatin group and MTX group were significantly lower than those in CIA group. Conclusion: Lovastatin can inhibit the levels of TNF-α, IL-6, IL-1β, IL-17 and IP-10.
