目的:观察复方芪苓无糖颗粒对高脂损伤血管内皮细胞的保护作用,探讨其治疗高脂血症的作用机制。方法:用高脂血清造成人血管内皮细胞(EVC)损伤模型,以复方芪苓无糖颗粒及血脂康含药血清进行干预。光镜下观察细胞形态变化,MTT法检测细胞活性;采用逆转录-聚合酶链反应(RT-PCR)技术检测复方芪苓无糖颗粒高、中、低剂量各组对高脂血清诱导的血管内皮细胞表面黏附分子(TGF-β1)基因表达。结果:复方芪苓无糖颗粒各剂量组及血脂康组均能使高脂损伤的EVC形态趋于正常,活性增强,细胞膜损伤减轻;并均有上调内皮细胞TGF-β1的趋势,血脂康组TGF-β1表达与复方芪苓颗粒中、高浓度组比较无统计学意义(P>0.05)。结论:复方芪苓无糖颗粒对高脂损伤的血管内皮细胞有保护作用,可能是其防治高脂血症及动脉粥样硬化的作用机制之一。
OBJECTIVE: To observe the protective effect of compound qi ling sugar-free granule on vascular endothelial cells of high fat injury and to explore its mechanism of action in treating hyperlipidemia. Methods: The model of human vascular endothelial cell (EVC) injury was induced by hyperlipidemic serum, and the compound qingling sugar-free granule and Xuezhikang serum were used for intervention. The morphological changes of cells were observed under light microscope, the cell viability was measured by MTT assay, and the effects of high, medium and low doses of compound QWL on the proliferation of vascular endothelial cells induced by hyperlipidemia were detected by reverse transcription-polymerase chain reaction (RT-PCR) Endothelial cell surface adhesion molecule (TGF-β1) gene expression. Results: The compound prescriptions of QLTG and Xuezhikang group all can make the morphology of EVC of hyperlipidemia tend to be normal, the activity is enhanced and the damage of cell membrane is alleviated. Both of them have the trend of up-regulating the expression of TGF-β1 in endothelial cells, The expression of TGF-β1 was not significantly different from the compound Qiliuling granule in high concentration group (P> 0.05). CONCLUSION: The compound QWL granule has a protective effect on vascular endothelial cells damaged by hyperlipidemia, which may be one of the mechanisms of its prevention and treatment of hyperlipidemia and atherosclerosis.
