目的建立测定小鼠血浆、心、肝、脾、肺、肾、脑中醋酸地塞米松含量的方法。方法色谱柱为Shimpack C18柱,流动相为甲醇-水(65:35),流速为1 ml.min-1,内标为甲睾酮,检测波长为240 nm。结果在生物样品中,醋酸地塞米松浓度1~100μg.g-1与峰面积比呈良好的线性关系,最低检测限为40 ng.g-1;方法回收率和RSD值均符合生物样品测定的要求。结论所建方法的线性、精密度、回收率均良好,且简便灵敏,可用于醋酸地塞米松制剂的药物动力学研究。
Objective To establish a method for determination of dexamethasone acetate in mouse plasma, heart, liver, spleen, lung, kidney and brain. Methods The chromatographic column was Shimpack C18 column. The mobile phase consisted of methanol-water (65:35) and flow rate was 1 ml.min-1. The internal standard was methyltestosterone and the detection wavelength was 240 nm. Results In biological samples, dexamethasone acetate concentration of 1 ~ 100μg.g-1 and peak area ratio showed a good linear relationship, the minimum detection limit of 40 ng.g-1; method recovery and RSD values are in line with biological samples Request. Conclusion The established method has good linearity, precision and recovery, and is simple and sensitive. It can be used to study the pharmacokinetics of dexamethasone acetate.
