目的:研究转录因子c-Jun及下游蛋白MMP-9在胃癌组织中的表达状况,分析其与临床病理特征和生存之间的关系。方法:应用免疫组化SP法,采用组织芯片技术,分析189例胃癌组织、54例癌旁组织、41例淋巴结转移灶及32例正常对照组织中c-Jun和MMP-9蛋白表达。结果:胃癌中c-Jun和MMP-9蛋白的阳性表达率分别为73.0%和78.3%。c-Jun蛋白的表达与分化程度有关(P<0.05);MMP-9的表达与浸润深度、淋巴结转移、分化程度及Lauren分型有关(P<0.05)。胃癌中c-Jun和MMP-9蛋白表达水平呈正相关(P<0.05)。41例淋巴结转移灶和相对应的41例胃癌组织各因子表达比较发现,MMP-9阳性表达率有显著性差异(P<0.05),c-Jun阳性表达率无显著性差异(P>0.05)。单因素生存分析显示c-Jun和MMP-9阳性组对胃癌患者生存期的影响有统计学意义(P<0.05),COX多元回归分析显示c-Jun和MMP-9的异常表达均不可作为影响胃癌预后的独立因素(P>0.05)。结论:c-Jun和MMP-9在胃癌组织中均有高表达。c-Jun阳性表达与胃癌的组织学分级密切相关,提示其异常表达可能是胃癌发生过程中的一个早期分子事件。MMP-9阳性表达与肿瘤的浸润、转移密切相关,可作为判断胃癌恶性行为的重要生物学标记物。c-Jun和MMP-9蛋白表达水平呈正相关。c-Jun和MMP-9蛋白阳性表达对于胃癌患者是一个不利的预后因素,尚不能作为一个独立的预后指标。
OBJECTIVE: To investigate the expression of c-Jun and MMP-9 in gastric carcinoma and to analyze its relationship with clinicopathological features and survival. Methods: The expressions of c-Jun and MMP-9 in 189 cases of gastric cancer tissues, 54 cases of paracancerous tissues, 41 cases of lymph node metastases and 32 cases of normal controls were analyzed by immunohistochemical SP method and tissue microarray technique. Results: The positive rates of c-Jun and MMP-9 in gastric cancer were 73.0% and 78.3% respectively. The expression of c-Jun protein was correlated with the degree of differentiation (P <0.05). The expression of MMP-9 was correlated with the depth of invasion, lymph node metastasis, differentiation and Lauren classification (P <0.05). The expression of c-Jun and MMP-9 in gastric cancer was positively correlated (P <0.05). The positive rate of MMP-9 was significantly different (P <0.05), while the positive rate of c-Jun had no significant difference (P> 0.05). There was no significant difference in the expression of MMP-9 among the 41 cases of lymph node metastasis and the corresponding 41 cases of gastric cancer . The univariate survival analysis showed that the c-Jun and MMP-9 positive groups had a significant effect on the survival of gastric cancer patients (P <0.05). Cox regression analysis showed that the abnormal expression of c-Jun and MMP-9 could not be affected Independent prognostic factors of gastric cancer (P> 0.05). Conclusion: Both c-Jun and MMP-9 are highly expressed in gastric cancer. The positive expression of c-Jun is closely related to the histological grade of gastric cancer, suggesting that its abnormal expression may be an early molecular event in the process of gastric cancer. The positive expression of MMP-9 is closely related to the infiltration and metastasis of the tumor, which can be used as an important biomarker to judge the malignant behavior of gastric cancer. There was a positive correlation between c-Jun and MMP-9 protein expression. The positive expression of c-Jun and MMP-9 protein is an unfavorable prognostic factor for gastric cancer patients and can not be used as an independent prognostic factor.
