测定红花药材及红花注射液中4种核苷类成分胞苷、尿苷、鸟苷和腺苷含量。采用Zorbax Eclipse XDB-C_(18)色谱柱;以0.05%三氟乙酸水溶液(A)和甲醇(B)为流动相梯度洗脱,在25℃条件下流速为1 mL/min;检测波长为260 nm。胞苷、尿苷、鸟苷、腺苷和尿嘧啶线性范围分别为4.02-603μg/mL(r~2=0.9998),9.38-1407μg/mL(r~2=0.9999),80.6-8060μg/mL(r~2=0.9999)及2.10-630μg/mL(r~2=0.9987);加样回收率分别为97.2%,94.5%,98.6%及108.5%。红花药材及红花注射液中4种核苷类成分的含量差异显著。本文首次报道了红花药材及红花注射液中核苷类成分的含量,其对完善红花药材及红花注射液的质量控制具有一定的指导意义。
The contents of cytidine, uridine, guanosine and adenosine in four kinds of nucleosides in Safflower and Safflower injection were determined. The mobile phase was eluted with Zorbax Eclipse XDB-C 18 column. The mobile phase consisted of 0.05% trifluoroacetic acid (A) and methanol (B) with a flow rate of 1 mL / min at 25 ℃. The detection wavelength was 260 nm. The linear ranges of cytidine, uridine, guanosine, adenosine and uracil were 4.02-603μg / mL (r 2 = 0.9998), 9.38-1407μg / mL (r 2 = 0.9999), 80.6-8060μg / mL r ~ 2 = 0.9999) and 2.10-630μg / mL (r ~ 2 = 0.9987) respectively. The recoveries were 97.2%, 94.5%, 98.6% and 108.5%, respectively. The contents of four nucleosides in safflower medicinal materials and safflower injection were significantly different. This article for the first time reported the safflower herbs and safflower injection nucleoside content, which has the guiding significance to improve the quality control of safflower herbs and safflower injection.
