本文扼要地介绍了暗场成象的基本原理和在核酸分子研究中的应用及其优越性。着重叙述和讨论了样品制备的主要环节和倾斜照明法的成象条件。应用暗场电镜技术和不同的制样方法研究了几种核酸和牛传染性鼻气管炎病毒(IBRV)的核衣壳,并报导了所取得的初步结果。
This article briefly introduces the basic principles of dark field imaging and its application in the study of nucleic acid molecules and their advantages. The main aspects of sample preparation and the imaging conditions of oblique illumination are mainly described and discussed. The nuclear capsids of several nucleic acids and bovine infectious rhinotracheitis virus (IBRV) were studied by dark field electron microscopy and different sample preparation methods, and the preliminary results obtained were reported.
