目的:应用Aβ1-42孵育PC12细胞株制作细胞损伤模型,探讨α-硫辛酸体外抗β-淀粉样蛋白(Aβ)损伤的作用。方法:用Aβ1-42和α-硫辛酸对PC12细胞进行处理,MTT法检测细胞活性,测定细胞培养上清液中LDH的活性检测细胞膜的损伤,测定细胞Caspase-3活性检测各组细胞的凋亡。结果:经Aβ1-42处理的PC12细胞,活性降低。用α-硫辛酸保护处理可提高细胞的存活率,降低LDH释放及Caspase-3活性,具有显著性差异(P<0.01)。结论:α-硫辛酸可拮抗Aβ1-42诱导的细胞损伤,对神经细胞具有一定的保护作用。
OBJECTIVE: To study the effect of α-lipoic acid on the anti-β-amyloid (Aβ) injury in vitro by using PC12 cells incubated with Aβ1-42. Methods: PC12 cells were treated with Aβ1-42 and α-lipoic acid. Cell viability was measured by MTT assay. LDH activity in cell culture supernatant was assayed for cell membrane damage. Caspase-3 activity was assayed for cell apoptosis Death. Results: Activity of PC12 cells treated with Aβ1-42 decreased. Treatment with α-lipoic acid increased cell viability and decreased LDH release and Caspase-3 activity (P <0.01). CONCLUSION: Alpha-lipoic acid can antagonize Aβ1-42-induced cell injury and has certain protective effect on nerve cells.
