目的:探讨神经生长因子(NGF)对离体培养在位子宫内膜基质细胞增殖及子宫腺肌病雌激素受体α表达的影响。方法:体外原代培养人子宫内膜细胞,用0 ng/ml、10 ng/ml、50 ng/ml和100 ng/ml的神经生长因子分别干预,用MTT法测定内膜基质细胞增殖情况,用Westen-blotting测定50 ng/ml NGF对基质细胞雌激素受体α表达的影响。结果:50 ng/ml的NGF可促进子宫内膜基质细胞增殖,细胞存活率为82.61%,比其他浓度NGF显著升高(P<0.05)。50 ng/ml NGF对病例组和对照组不同细胞雌激素受体表达有影响,病例组腺上皮细胞和基质细胞总体均数差异均具有统计学意义(P<0.05)。结论:50 ng/ml的NGF可促进在位内膜基质细胞增殖及子宫腺肌病腺上皮细胞和基质细胞雌激素受体α表达。
Objective: To investigate the effect of nerve growth factor (NGF) on estrogen receptor α expression in cultured endometrial stromal cells and adenomyosis in vitro. Methods: Human endometrial cells were primarily cultured in vitro. The cells were treated with 0 ng / ml, 10 ng / ml, 50 ng / ml and 100 ng / ml NGF respectively. The proliferation of endometrial stromal cells was measured by MTT assay. The effect of 50 ng / ml NGF on estrogen receptor α expression in stromal cells was determined by Westen-blotting. Results: NGF (50 ng / ml) could promote the proliferation of endometrial stromal cells with a cell viability of 82.61%, which was significantly higher than that of other concentrations of NGF (P <0.05). The expression of estrogen receptor in different cell groups was significantly different from that in 50 ng / ml NGF group (P <0.05). Conclusion: NGF (50 ng / ml) can promote the proliferation of eutopic endometrial stromal cells and the expression of estrogen receptor α in glandular epithelial cells and stromal cells of adenomyosis.
