目的:利用介孔二氧化硅纳米颗粒负载顺铂,并研究对人舌鳞状细胞癌细胞(TSCCs)的杀伤作用。方法:以表面活性剂CTAB为模板合成MCM-41介孔二氧化硅纳米颗粒(Mesoporous Silica Nanoparticle,MSN),并负载顺铂,通过扫描电镜和红外光谱观察颗粒表面形貌和功能团组成,通过碘化丙啶(PI)染色法观察TSCCs的细胞凋亡。结果:MSN平均粒径约200 nm,细胞结果显示载药纳米颗粒组死亡细胞明显多于其它各组(P<0.01),且单纯纳米颗粒组死亡细胞低于空白对照组。结论:MSN负载顺铂后显著增强了对肿瘤细胞的杀伤能力,有望应用于临床肿瘤化疗提高药效或降低药物并发症。
OBJECTIVE: To use mesoporous silica nanoparticles to support cisplatin and study its cytotoxicity on human tongue squamous cell carcinoma (TSCCs) cells. METHODS: MCM-41 mesoporous silica nanoparticles (MSN) were synthesized using surfactant CTAB as a template and loaded with cisplatin. The surface morphology and functional group composition of the particles were observed by scanning electron microscopy and infrared spectroscopy. Propidium iodide (PI) staining was used to observe the apoptosis of TSCCs. Results: The mean particle size of MSN was about 200 nm. The results of cell assay showed that the number of dead cells in the drug-loaded nanoparticles group was significantly more than that in other groups (P <0.01), and the number of dead cells in the nanoparticle-only group was lower than that in the blank control group. Conclusion: MSN loaded with cisplatin significantly enhanced the killing ability of tumor cells, and is expected to be used in clinical tumor chemotherapy to improve efficacy or reduce drug complications.
