目的探讨甲羟孕酮(MPA)及米非司酮(RU486)对人卵巢癌耐药细胞系SKOV-3/DDP耐药性的影响。方法2005-05-2006-03对吉林大学第二医院建立的人卵巢癌顺铂(DDP)耐药细胞株SKOV-3/DDP采用四甲基偶氮唑蓝(MTT)法,比较不同质量浓度MPA、RU486、甲羟孕酮(MPA)+RU486对SKOV-3/DDP细胞生长及对DDP耐药性的影响。结果20mg/L以上RU486或64mg/LMPA能够抑制SKOV-3/DDP细胞增殖;≥64mg/LMPA或≥20mg/LRU486能够增强该细胞系对DDP的敏感性(P<0.05)。RU486质量浓度为40mg/L时,MPA+RU486对SKOV-3/DDP细胞生长有抑制作用,可以逆转DDP引发的耐药。在同一MPA质量浓度下,随着DDP质量浓度的升高,抑制率逐渐升高。结论较大剂量MPA或RU486可以逆转DDP引发的耐药;MPA、RU486合用可以增强DDP的作用。
Objective To investigate the effects of medroxyprogesterone (MPA) and mifepristone (RU486) on the drug resistance of human ovarian cancer cell line SKOV-3 / DDP. METHODS: The human ovarian cancer cell line DDP resistant to SKOV-3 / DDP established in the Second Hospital of Jilin University was studied by MTT assay. The effects of different concentrations of Effects of MPA, RU486, MPA + RU486 on the Cell Growth and Drug Resistance of SKOV-3 / DDP Cells. Results RU486 or 64mg / LMPA above 20mg / L could inhibit the proliferation of SKOV-3 / DDP cells; and ≥64mg / LMPA or ≥20mg / LRU486 could enhance the sensitivity of this cell line to DDP (P <0.05). At RU486 concentration of 40 mg / L, MPA + RU486 could inhibit the growth of SKOV-3 / DDP cells and reverse the drug resistance induced by DDP. At the same concentration of MPA, with the increase of DDP mass concentration, the inhibition rate gradually increased. Conclusions Higher doses of MPA or RU486 can reverse the drug resistance induced by DDP. Combination of MPA and RU486 can enhance the effect of DDP.
