目的研究中药有效成分人参皂甙Rg1、肉桂酸和丹参酮ⅡA组合(RCT)对人成骨肉瘤MG-63细胞的形态与超微结构改变和终末分化的影响,鉴定其对人成骨肉瘤细胞终末分化的诱导作用。方法以中药有效成分组合(33mg/L人参皂甙Rg1+2mmol/L肉桂酸+0.3mg/L丹参酮IIA)处理MG-63细胞7d,对照组与处理组样本各3次重复,免疫细胞化学检测及光镜与电镜观察系统研究RCT组合对MG-63细胞的细胞生物学效应。环六亚甲基双乙酰胺(HMBA)处理MG-63细胞作为分化诱导效果的阳性对照。结果免疫细胞化学检测显示,成骨肉瘤细胞终末分化指标Ⅰ型胶原、骨黏素、骨钙蛋白的表达呈强阳性,并观察到钙化糖原颗粒增多与典型骨结节的形成;光镜与电镜观察结果显示,处理后的MG-63细胞产生了细胞形态规则,大小一致,细胞铺展体积增大,核质比例减小,核内核仁数目减少,细胞器丰富发达等趋向正常细胞特征的显著改变。结论RCT组合能显著改变MG-63细胞形态与超微结构恶性特征,并促进终末分化标志物的表达,从而对人成骨肉瘤MG-63细胞的终末分化具有显著诱导作用。

Conclusion RCT can significantly change the morphological and ultrastructural features of MG-63 cells and promote the expression of terminal differentiation markers, which can induce the terminal differentiation of human osteosarcoma MG-63 cells.
