观察尾叶香茶菜中提取物二萜类化合物F对人肝癌细胞株SMMC-7721的作用并探讨其机制。分别应用MTT法、AO/EB荧光染色法分析细胞存活率和凋亡情况,利用DCFH-DA荧光分光光度法检测细胞内过氧化物(ROS)的水平。二萜类化合物F抑制SMMC-7721细胞增殖,且呈时间剂量依赖性,二萜类化合物F作用的SMMC-7721细胞内ROS水平明显升高。二萜类化合物F可以启动细胞内氧化应激诱导SMMC-7721细胞坏死或凋亡而抑制肝癌细胞株增殖。
To observe the effect of diterpenoid F, an extract from Chayao japonica, on human hepatocellular carcinoma cell line SMMC-7721 and its mechanism. Cell viability and apoptosis were analyzed by MTT assay and AO / EB fluorescence staining, and the level of intracellular peroxide (ROS) was detected by DCFH-DA fluorescence spectrophotometry. Diterpenoid F inhibited the proliferation of SMMC-7721 cells in a time-and dose-dependent manner. The level of ROS in SMMC-7721 cells treated with diterpenoid F was significantly increased. Diterpenoid F can inhibit the proliferation of hepatocellular carcinoma cells by inducing intracellular oxidative stress-induced necrosis or apoptosis of SMMC-7721 cells.
