目的探讨中国山东汉族人群肿瘤坏死因子凋亡相关诱导配体(Tumor necrosis factor(TNF)-related apoptosis in-ducing ligand,TRAIL)基因启动子区单核苷酸多态性(single nucleotide polymorphism,SNP)与脂肪肝(FLD)的相关性。方法采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)分析和四引物法聚合酶链反应(Tetra-primer ARMS-PCR)的方法检测了42例非酒精性脂肪肝6、7例酒精性脂肪肝和132例健康对照者的TRAIL基因启动子区单核苷酸多态性位点-707C/T、-665T/C、-621C/T和-597A/G的基因型,计算相应的基因型频率及等位基因频率,进行χ2检验。结果-707C/T、-665T/C和-597A/G位点在中国山东汉族人群中未检测到单核苷酸多态性;-621C/T位点在中国山东汉族人群中存在单核苷酸多态性,且该位点CC/CT/TT基因型频率在非酒精性脂肪肝、酒精性脂肪肝以及正常对照组中分别为0.238/0.500/0.262、0.269/0.448/0.284和0.280/0.394/0.326。但该位点的基因型频率和等位基因频率在三组之间均没有统计学差异。结论在中国山东汉族人群中,仅检测到TRAIL基因启动子区-621C/T位点的单核苷酸多态性,该多态性与脂肪肝的易感性没有统计学差异(P>0.05)。
Objective To investigate the single nucleotide polymorphisms (SNPs) in the promoter region of tumor necrosis factor (TNF) -related apoptosis in-ducing ligand (TRAIL) Correlation with fatty liver (FLD). Methods Forty-two patients with non-alcoholic fatty liver were examined by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and Tetra-primer ARMS-PCR Cases of alcoholic fatty liver and 132 healthy controls TRAIL gene promoter region single nucleotide polymorphism sites -707C / T, -665T / C, -621C / T and -597A / G genotypes, calculated The corresponding genotype frequency and allele frequency, χ2 test. Results No single nucleotide polymorphisms were detected in -707C / T, -665T / C and -597A / G loci in Shandong Han population in China. The presence of mononucleoside at -621C / T locus The polymorphisms of CC / CT / TT genotypes were 0.238 / 0.500 / 0.262, 0.269 / 0.448 / 0.284 and 0.280 / 0.394 in non-alcoholic fatty liver, alcoholic fatty liver and normal control group respectively /0.326. However, there was no significant difference in genotype frequency and allele frequency between the three groups. Conclusion Only the single nucleotide polymorphism (SNP) of -621C / T locus in the promoter region of TRAIL gene was detected in Shandong Han Chinese population. There was no significant difference between the polymorphism and fatty liver disease (P> 0.05) .
