用MTT比色分析法和流式细胞术在体外研究了5肽胃泌素(pentagastrin,PG)及胃泌素受体拮抗剂丙谷胺(proglumide,PGL)对人大肠癌细胞株SW480的作用。结果显示:PG能促进SW480细胞生长;PGL单用时对SW480细胞没有影响,当PG与PGL合用时,PGL呈剂量依赖地拮抗PG的作用,PGL在32μg/ml以上时,拮抗作用趋于恒定。提示胃泌素的作用是通过其受体发挥的。PG促进SW480细胞合成DNA、蛋白质,促进SW480细胞从G0/G1期过度到S期和G2M期;PGL完全拮抗这种作用,提示胃泌素的作用在受体后是通过影响细胞周期实现的
The effects of pentagastrin (PG) and gastrin receptor antagonist proglumide (PGL) on human colorectal cancer cell line SW480 were studied in vitro by MTT colorimetric assay and flow cytometry. . The results showed that PG could promote the growth of SW480 cells; PGL alone had no effect on SW480 cells. When PG was combined with PGL, PGL antagonized the effect of PG in a dose-dependent manner. When PGL was above 32 μg/ml, the antagonism tended to be constant. It is suggested that the effect of gastrin is exerted through its receptor. PG promotes the synthesis of DNA and protein in SW480 cells and promotes the transition from G0/G1 phase to S phase and G2M phase in SW480 cells; PGL completely antagonizes this effect, suggesting that the effect of gastrin on the receptor is through affecting the cell cycle.
