本试验用化学分析及聚丙烯酰胺凝胶电泳方法,分析和分离了小地老虎[Agrotis ypsilon(Rottemberg)]在个体发育期间血淋巴蛋白的含量和类型变动的情况,用以说明夜蛾科昆虫的变态特征,以及有关越冬、迁飞、滞育等的生理基础。初步试验的结果表明:(1)血淋巴蛋白的含量,从五龄幼虫末期的1.21克/100毫升增加到蛹前期(蛹龄四天)的5.60克/100毫升,即可增加4.6倍左右。而在五龄幼虫变六龄,预蛹变蛹以及蛹羽化之前,即在每次蜕皮期间,血淋巴蛋白的含量都明显下降。(2)用聚丙烯酰胺凝胶电泳分离血淋巴蛋白所得的谱带表明,幼虫期至少存在有12条谱带,蛹期有17条,至成虫期,谱带明显减少(主要蛋白带,雄蛾3条,雌蛾4条)。同时还发现,蛹期的特征性谱带是与幼虫期蜕皮时的谱带相一致的。从各地区收集的卵孵化出来的幼虫,经过饲育到蛹,它们的谱带也无差异。(3)雌蛾的特异蛋白带,一般需在羽化后三天才出现,并与卵巢管内卵黄沉积较迟相符合,说明小地老虎雌蛾在迁飞前有一个短暂的生殖滞育期。
This experiment analyzed and isolated the changes of hemolymph protein content and type during the development of the individual Agrotis ypsilon (Rottemberg) by chemical analysis and polyacrylamide gel electrophoresis to illustrate that the noctuid insects Metamorphosis, as well as on the physiological basis of overwintering, migrating, diapause. Preliminary results showed that: (1) The content of hemolymph protein increased from 4.6 g / 100 ml at 1.21 g / 100 ml at the end of fifth instar larvae to 5.60 g / 100 ml at pubertal (four days old). By the adult stage, the band was significantly reduced (main protein band, male 3 moths, 4 female moths). At the same time, it was also found that the characteristic bands of pupae are consistent with those of the larval stage molting. The larvae that hatch from the eggs collected from all regions, after breeding to pupae, showed no difference in their bands. (3) The specific protein band of the female moth usually appeared three days after eclosion and was consistent with the late deposition of the egg yolk in the ovary tube, indicating that the female Timberlake had a short reproductive diapause period before its migration.
