目的:探讨补肾益髓中药含药血清、转化生长因子-β1(TGF-β1)、骨形态发生蛋白2(BMP-2)联合诱导大鼠骨髓间充质干细胞(BMSCs)增殖、骨向分化能力的研究。方法:采用全骨髓贴壁法分离及培养大鼠BMSCs。补肾益髓中药含药血清、TGF-β1及BMP-2的制备并确定最佳促BMSCs增殖、骨向分化添加浓度。实验分组:空白组、血清组、TGF-β1组、血清+TGF-β1组、BMP-2组、血清+BMP-2组、TGF-β1+BMP-2组、综合诱导组,酶联免疫分析检测Coll、BGP浓度。结果:TGF-β1最佳促BMSCs增殖浓度为10 ng/m L,BMP-2最佳促BMSCs骨向分化浓度是40 ng/m L;综合诱导组能明显促进大鼠BMSCs的I型胶原(Coll)、骨钙素(BGP)的表达量。结论:中西医联合诱导能够显著提高大鼠BMSCs增殖、骨向分化能力。
Objective: To investigate the effects of Bushen Yisui Chinese medicinal serum, transforming growth factor-β1 (TGF-β1) and bone morphogenetic protein 2 (BMP-2) on the proliferation and differentiation of rat bone marrow mesenchymal stem cells Research. Methods: BMSCs were isolated and cultured using whole bone marrow adherence method. Bushenyishan medicine containing serum, TGF-β1 and BMP-2 preparation and to determine the best promote BMSCs proliferation, bone differentiation to add concentration. The experimental groups were as follows: blank group, serum group, TGF-β1 group, serum + TGF-β1 group, BMP-2 group, serum + BMP-2 group, TGF-β1 + BMP-2 group, Coll and BGP concentration detection. RESULTS: The optimal concentration of TGF-β1 to promote BMSCs was 10 ng / m L, and the best differentiation rate of BMP-2 to BMSCs was 40 ng / m L. The induction group could significantly promote type I collagen Coll) and osteocalcin (BGP). Conclusion: The combination of TCM and WM can significantly improve the proliferation and differentiation of BMSCs in rats.
