目的:观察白芍总苷(TGP)对类风湿关节炎(RA)患者外周血中T淋巴细胞表达破骨细胞分化因子(RANKL)的影响。方法:采用流式细胞术检测RA患者及健康志愿者T细胞表达RANKL情况;观察白芍总苷对RA患者T细胞表达RANKL的影响。结果:与健康志愿者组相比,活动期RA患者T细胞RANKL表达率显著增高(P<0.01);体外培养的RA患者外周血细胞被白芍总苷50、100ug/ml剂量作用,可使T细胞RANKL表达量明显下降(P<0.05,P<0.01)。结论:白芍总苷可通过抑制T细胞表达RANKL,从而减慢由RANKL介导的破骨细胞活化及骨质破坏。
Objective: To observe the effect of TGP on the expression of osteoclast differentiation factor (RANKL) in peripheral blood of patients with rheumatoid arthritis (RA). Methods: The expression of RANKL in T cells of RA patients and healthy volunteers was detected by flow cytometry. The effect of total glucosides of peony on RANKL expression in T cells of RA patients was observed. Results: Compared with healthy volunteers, the expression of RANKL was significantly increased in patients with active RA (P <0.01). The peripheral blood cells of RA patients were treated with 50,100ug / ml of total glucosides of Paeony, Cell RANKL expression decreased significantly (P <0.05, P <0.01). CONCLUSION: Total glucosides of paeony can inhibit RANKL-mediated osteoclast activation and bone destruction by inhibiting the expression of RANKL in T cells.
