【目的】探讨骨碎补总黄酮对家兔股骨牵张成骨的影响。【方法】选用健康家兔32只,随机分为治疗组和对照组,每组16只。行单侧股骨牵张术,以自制延长器固定。经7 d延迟期,以1 mm/d的速度牵张,2次/d,连续10 d。治疗组动物自术后第1天用骨碎补总黄酮溶液灌胃,至实验结束。固定28 d后处死动物,留取标本,行组织学观察及免疫组织化学检测。【结果】治疗组新生骨组织成熟骨质区域增大,成骨细胞数、骨小梁面积百分比均显著高于对照组,骨形态发生蛋白-2(BMP-2)和转化生长因子-β1(TGF-β1)呈棕色深染,染色程度显著高于对照组。【结论】骨碎补总黄酮能有效地加速骨牵张中新生骨质的生成与成熟。
【Objective】 To investigate the effects of total flavonoids of Rhizoma Drynariae on femoral distraction osteogenesis in rabbits. 【Methods】 Thirty-two healthy rabbits were randomly divided into treatment group and control group, with 16 in each group. Unilateral femoral distraction surgery, self-made extender fixation. After 7 d delay period, 1 mm / d speed stretch, 2 times / d, continuous 10 d. The animals in the treatment group were orally gavaged with total flavonoids of Rhizoma Drynariae on the first day after operation until the end of the experiment. After 28 days, the animals were sacrificed and the specimens were taken for histological observation and immunohistochemistry. 【Results】 The bone mineral density and osteoblast area in the treated group were significantly higher than those in the control group. The bone morphogenetic protein-2 (BMP-2) and transforming growth factor-β1 TGF-β1) was stained dark brown, staining was significantly higher than the control group. 【Conclusion】 Total Flavonoids of Rhizoma Drynariae can effectively accelerate the formation and maturation of new bone in bone distraction.
