目的 探讨Bitl蛋白在膀胱癌中的表达以及临床意义.方法 采取Real-time PCR和Westernblot法分别检测Bit1mRNA和蛋白含量在正常膀胱组织细胞系SV及膀胱癌细胞系T24和5637中的表达.通过RNA干扰技术敲除Bit1,进一步检测Bit1mRNA和蛋白含量以判断敲除效力,通过MTT实验检测各细胞系的细胞增殖能力.结果 Bit1mRNA和蛋白表达水平在膀胱癌细胞系T24和5637中明显高于SV细胞系(P<0.05),转染Si-RNA后Bit1mRNA和蛋白含量显著降低.MTT的结果显示转染Si-RNA后T24和5637细胞系的细胞增殖能力均显著降低.结论 Bit1在膀胱癌中高表达,在膀胱癌发生发展过程中发挥重要作用,可能是一种潜在的肿瘤标记物.“,”Objective To investigate the expression and clinical significance of Bitl mRNA and protein in the bladder cancer.Methods Real-time PCR and Western blot were used to detect the expressions of Bit1 mRNA and protein in SV,T24 and 5637 cell lines.By knocking down Bit1 with RNA interference,the expressions of Bit1 mRNA and protein were examined to determine the knockdown efficacy.MTT analysis was used to test the cell proliferation ability of each cell line.Results The expression levels of Bit1 mRNA and protein were significantly higher in T24 and 5637 cell lines than in SV cell line(P<0.05),but decreased significantly after transfection of Si-RNA.MTT analysis showed that the proliferation ability of T24 and 5637 cell lines also decreased significantly after transfection with Si-Bitl.Conclusion Bit1 might be involved in the pathogenesis of bladder cancer and a potential tumor marker of bladder cancer at the early stage.
