目的 :探讨血管内皮生长因子 (vascular endothelial growth factor ,VEGF)的表达、血管生成与骨肉瘤的关系 ,为判断骨肉瘤的恶性程度及预后提供新的指标。方法 :应用免疫组化技术分别对 46例骨肉瘤标本 (经过Price分级 )进行 VEGF和微血管密度 (m icrovessel density ,MVD)染色 ,研究 VEGF表达、微血管密度之间及其与骨肉瘤的关系。结果 :骨肉瘤标本中 Price 级与 级、 级与 级之间的 VEGF表达有显著性差异 (P <0 .0 1)。Price 、 级的 VEGF表达强度高于 Price 级的表达强度 (P <0 .0 5 )。骨肉瘤标本中 Price各级之间的 MVD有显著性差异 ,且 MVD从 Price 级到 级呈递增趋势 ,微血管密度随着 Price分级的增加而逐渐增高 (P<0 .0 5 )。VEGF表达阳性组的骨肉瘤标本 MVD明显高于 VEGF表达阴性组 ,且随着 VEGF表达的增强 ,微血管密度随之增加 (P <0 .0 5 )。结论 :分化程度低、恶性程度高的骨肉瘤 VEGF表达增强 ;骨肉瘤的分化程度越低、恶性程度越高MVD越高 ;VEGF与骨肉瘤的血管生成密切相关 ,VEGF表达增强 ,微血管密度增高 ;VEGF及 MVD可能作为判定骨肉瘤预后的参考。
Objective: To investigate the relationship between the expression of vascular endothelial growth factor (VEGF) and angiogenesis and osteosarcoma, and to provide a new index for judging the malignancy and prognosis of osteosarcoma. METHODS: Immunohistochemical technique was used to quantify VEGF and micro-vessel density (MVD) in 46 cases of osteosarcoma specimens (after price classification) to study the relationship between VEGF expression, microvessel density, and osteosarcoma. Results: There was a significant difference in the expression of VEGF between the grades and grades and grades in osteosarcoma specimens (P < 0.01). The expression level of VEGF at the price and grade was higher than that at the price level (P < 0.05). There was a significant difference in the MVD between the various levels of osteosarcoma specimens, and the MVD showed an increasing trend from the grade level to the grade, and the microvessel density gradually increased with the increase of the price grade (P<0.05). The MVD of osteosarcoma specimens with positive VEGF expression was significantly higher than that with negative VEGF expression. With the increase of VEGF expression, microvessel density increased (P < 0.05). Conclusion: The expression of VEGF in osteosarcoma with low degree of differentiation and high degree of malignancy is enhanced. The lower the degree of differentiation of osteosarcoma, the higher the degree of malignancy is, the higher the MVD is; VEGF is closely related to the angiogenesis of osteosarcoma; the expression of VEGF is increased and the microvessel density is increased; VEGF and MVD may serve as a reference for the prognosis of osteosarcoma.
