目的筛选米碎花Eurya chinensis茎叶的微小核糖核酸(Micro RNAs,mi RNAs)类药效物质及其靶基因功能分析。方法采用高通量测序法,测定米碎花、正常小鼠和喂食米碎花匀浆液小鼠肝组织及血浆中mi RNAs的表达丰度,并与Gene Bank上的植物mi RNAs数据库比对,结合实时荧光定量PCR(RTFQ-PCR)验证,鉴定吸收进入体内的米碎花mi RNAs。以人类m RNA为靶标,采用Target Scan与Mi Randa预测米碎花mi RNAs的靶基因,并通过基因功能聚类(GO)与KEGG分析靶基因的相关功能。结果共鉴定出5个米碎花mi RNAs可吸收进入体内,其主要在生化过程、细胞成分和分子功能3大区域发挥作用,并影响肿瘤、代谢、炎症等相关信号通路。结论 mi RNAs可能是一类新型的中药药效物质。
Objective To screen the microRNAs (mi RNAs) functional substances in stems and leaves of Mica chrysanthemum and analyze the function of their target genes. Methods The high-throughput sequencing method was used to determine the abundance of mi RNAs in liver tissues and plasma of Mice flower, normal mice and homogenates of mice fed with Mice, and compared with the miRNAs database of Gene Bank. Combined with real-time fluorescence quantitative PCR (RTFQ-PCR) validation, identified mi floral mi RNAs absorbed into the body. Using human m RNA as a target, Target Scan and Mi Randa were used to predict the target genes of rice miRNAs and their functions were analyzed by gene function clustering (GO) and KEGG. Results A total of 5 rice miRNAs were identified and could be absorbed into the body. These miRNAs mainly function in three regions of biochemical processes, cellular components and molecular functions, and affect the related signaling pathways such as tumor, metabolism and inflammation. Conclusion mi RNAs may be a new type of traditional Chinese medicine.
