目的建立一种高灵敏度的液质联用方法,用于快速测定大鼠血浆中天麻素的浓度,并计算了天麻素在大鼠体内的药代动力学参数。方法血浆样品加乙腈沉淀蛋白,以乙腈-水(68:32)为流动相,用ApolloC18色谱柱(4.6mm×150mm,5μm)分离,采用电喷雾离子化源(ESI)三重四级杆串联质谱,以选择离子监测(SIM)方式进行检测。结果天麻素线性范围为:0.0222~2.22μg·mL-1,回归方程为:A=301733+2×107C,R2=0.9949(n=7);低、中、高3个质量浓度的平均回收率分别为:99.55%、99.10%、98.65%。结论该法快速、灵敏、专属,适用于测定生物样本中天麻素的血药浓度。
