目的研究hOGG1、XRCC1、XRCC3单核苷酸多态性与食管癌放射敏感性的关系。方法采用PCR-RFLP法检测94例食管鳞状细胞癌患者外周血hOGG1、XRCC1、XRCC3基因单核苷酸多态性,并分析hOGG1 Ser326Cys、XRCC1 Arg399Gln、XRCC3 Thr241Met单核苷酸多态性在食管癌放射治疗有效组及放射治疗无效组中的分布情况。结果 94例食管鳞癌患者经根治性三维适形或调强放疗,有效率为84.06%,XRCC1Arg399Gln等位基因频率在食管癌放射治疗有效组及无效组中的分布差异有统计学意义(P=0.001)。hOGG1Ser326Cys及XRCC3 Thr241Met等位基因在两组中的分布差异均无统计学意义(P=0.586、0.152)。XRCC1Arg/Arg、Arg/Gln、Gln/Gln基因型患者放疗有效率分别为91.5%、88.9%和60.0%,3种基因型放疗有效率差异有统计学意义(P=0.009)。hOGG1 Ser326Cys及XRCC3 Thr241Met各基因型间放疗有效率差异无统计学意义(P>0.05)。结论 XRCC3 Arg399Gln单核苷酸多态性与食管癌放射敏感性存在相关性;hOGG1 Ser326Cys,XRCC1 Thr241Met基因多态性与食管癌放射敏感性无明显相关性。
Objective To investigate the relationship between single nucleotide polymorphisms of hOGG1, XRCC1 and XRCC3 and radiosensitivity of esophageal cancer. Methods The single nucleotide polymorphisms of hOGG1, XRCC1 and XRCC3 in peripheral blood of 94 patients with esophageal squamous cell carcinoma were detected by PCR-RFLP. The single nucleotide polymorphisms of hOGG1 Ser326Cys, XRCC1 Arg399Gln and XRCC3 Thr241Met were detected in the esophagus Cancer radiotherapy effective group and radiotherapy invalid group distribution. Results 94 patients with esophageal squamous cell carcinoma by radical three-dimensional conformal or intensity modulated radiotherapy, the effective rate was 84.06%, XRCC1Arg399Gln allele frequency in esophageal cancer radiotherapy effective group and ineffective group, the distribution was statistically significant (P = 0.001). There were no significant differences in the distribution of hOGG1Ser326Cys and XRCC3 Thr241Met alleles between the two groups (P = 0.586,0.152). The effective rate of radiotherapy in patients with XRCC1Arg / Arg, Arg / Gln and Gln / Gln genotypes was 91.5%, 88.9% and 60.0%, respectively. There was significant difference in radiotherapy efficiency between the three genotypes (P = 0.009). There was no significant difference in the effective rate of radiotherapy between hOGG1 Ser326Cys and XRCC3 Thr241Met genotypes (P> 0.05). Conclusion There is a correlation between XRCC3 Arg399Gln SNP and esophageal cancer radiosensitivity. The polymorphisms of hOGG1 Ser326Cys and XRCC1 Thr241Met have no significant correlation with esophageal cancer radiosensitivity.
