目的:检测上皮性卵巢癌中特异AT序列结合蛋白1(Special AT-rich sequence-binding protein 1,SATB1)的表达,探讨SATB1表达与卵巢癌的发生,进展及转移的关系及意义。方法:以实时荧光定量RT-PCR和免疫组化方法检测91例上皮性卵巢癌组织、13例卵巢交界性囊腺瘤及8例正常卵巢组织中SATB1mRNA和蛋白的表达,分析SATB1基因表达与临床病理参数的相关性。结果:上皮性卵巢癌组织和交界性囊腺瘤组织中SATB1mRNA表达量分别为正常卵巢组织的6.97倍和5.70倍,差异有统计学意义(P<0.05)。SATB1mRNA表达水平随着卵巢癌分期升高而增加:Ⅰ期,Ⅱ期和Ⅲ期分别为正常卵巢组织的5.31倍,6.67倍和8.04倍(P<0.05),并且在淋巴结转移组表达明显高于无转移组(P<0.05)。免疫组化也显示SATB1蛋白在上皮性卵巢癌中表达高于正常卵巢组织,差异具有统计学意义(P<0.05)。结论:SATB1的表达水平可能与上皮性卵巢癌发生,进展及转移有关。
OBJECTIVE: To detect the expression of special AT-rich sequence-binding protein 1 (SATB1) in epithelial ovarian cancer and to explore the relationship between SATB1 expression and the occurrence, progression and metastasis of ovarian cancer. Methods: The expression of SATB1 mRNA and protein in 91 cases of epithelial ovarian cancer, 13 cases of ovarian borderline cystadenoma and 8 cases of normal ovarian tissue were detected by real-time fluorescence quantitative RT-PCR and immunohistochemistry. The expressions of SATB1 gene and clinical Correlation of pathological parameters. Results: The expressions of SATB1mRNA in epithelial ovarian cancer and borderline cystadenoma were 6.97 folds and 5.70 folds higher than those in normal ovarian tissues, respectively (P <0.05). The expression of SATB1mRNA increased with the stage of ovarian cancer staging: stage Ⅰ, stage Ⅱ and stage Ⅲ were 5.31 times, 6.67 times and 8.04 times higher than that of normal ovarian tissue respectively (P <0.05), and the expression of SATB1 mRNA was significantly higher in lymph node metastasis group No metastasis (P <0.05). Immunohistochemistry also showed that SATB1 protein was higher in epithelial ovarian cancer than in normal ovarian tissue, the difference was statistically significant (P <0.05). Conclusion: The expression of SATB1 may be related to the occurrence, progression and metastasis of epithelial ovarian cancer.
