目的探讨粒/巨噬细胞集落刺激因子(GM-CSF)及白介素-4(IL-4)对正常成人及大肠癌患者树突状细胞(DC)表面人白细胞抗原(HLA)-DR及B7等免疫分子表达及其免疫功能的影响。方法分离正常成人(n=10)及大肠癌患者(n=10)外周血DC,以GM-CSF及IL-4联合刺激正常成人及大肠癌者DC,检测GM-CSF及IL-4联合刺激前后DC表面HLA-DR及B7表达水平及DC免疫功能变化。结果经GM-CSF及IL-4联合刺激后,大肠癌患者DC表面HLA-DR及B7表达水平由9.2±1.4、8.6±1.5增至17.2±1.2、15±1.3VOF(P<0.01),DC免疫诱导能力由6356±112增至1042±146cpm(P<0.01);正常成人DC表面HLA-DR及B7表达水平由15±1.6、16±0.9增至18±1.5、18±1.3VOF(P>0.05),DC免疫诱导能力由9806±113增至1108±127cpm(P>0.05)。结论GM-CSF及IL-4联合刺激能增强正常成人及大肠癌患者DC表面HLA-DR及B7表达水平并进一步增强DC免疫功能。提示联合应用GM-CSF及IL-4可能成为提高宿主抗?
Objective To investigate the effects of granulocyte-macrophage colony-stimulating factor (GM-CSF) and interleukin-4 (IL-4) on the surface of dendritic cells (DCs) in normal adult and colorectal cancer patients. Human leukocyte antigen (HLA)-DR and B7, etc. The expression of immune molecules and the impact of their immune function. Methods Peripheral blood DC was isolated from normal adults (n=10) and colorectal cancer patients (n=10). Normal and adult colorectal DCs were stimulated with GM-CSF and IL-4 to detect the combined stimulation of GM-CSF and IL-4. Changes of HLA-DR and B7 expression and DC immune function in DC surface before and after. Results After combined stimulation of GM-CSF and IL-4, the expression levels of HLA-DR and B7 on DC surface of colorectal cancer patients increased from 9.2±1.4, 8.6±1.5 to 17.2±1.2. At 15±1.3 VOF (P<0.01), the immune induction ability of DC increased from 6356±112 to 1042±146 cpm (P<0.01); the level of HLA-DR and B7 expression on normal adult DC surface was 15±1. The levels of .6 and 16±0.9 increased to 18±1.5, 18±1.3 VOF (P>0.05), and the immune induction ability of DC increased from 9806±113 to 1108±127 cpm (P>0.05). Conclusion The combined stimulation of GM-CSF and IL-4 can enhance the expression of HLA-DR and B7 on the surface of normal adult and colorectal cancer DCs, and further enhance the DC immune function. It suggests that the combined use of GM-CSF and IL-4 may be used to increase host resistance.
