目的 :观察溃疡性结肠炎 (UC)患者周围血中性粒细胞 (PMN)凋亡的变化 ,并检测周围血中细胞因子IL 8、IL 6、TNF α、NO水平的变化 ,探讨细胞因子对PMN凋亡的影响。方法 :采用流式细胞术检测 2 8例UC病人周围血PMN凋亡率。结果 :活动期UC患者PMN凋亡率明显低于健康人对照组和缓解期UC患者。缓解期UC患者PMN凋亡率与对照组无差别。不同病情活动期UC患者之间PMN凋亡率有显著性差异。活动期UC患者周围血中IL 8、IL 6、TNF α、NO水平均高于对照组和缓解组 ,且与PMN凋亡呈负相关 ,与病情呈正相关。缓解组患者上述细胞因子与对照组无显著性差异。结论 :UC患者PMN凋亡延迟 ,且与病情及疗效密切相关。各种炎性细胞因子产生过多可能是导致PMN凋亡延迟的重要机制
Objective: To observe the changes of apoptosis of peripheral blood neutrophil (PMN) in patients with ulcerative colitis (UC) and to detect the changes of cytokines IL 8, IL 6, TNF α and NO in peripheral blood, Effect of PMN apoptosis. Methods: Flow cytometry was used to detect the apoptosis rate of peripheral blood PMN in 28 patients with UC. Results: The apoptosis rate of PMN in active UC patients was significantly lower than that in healthy controls and remission UC patients. The PMN apoptosis rate in UC patients was similar to that in control group. There was a significant difference in the apoptotic rate of PMN between UC patients with different stages of disease activity. The levels of IL 8, IL 6, TNF α and NO in the peripheral blood of patients with active UC were significantly higher than those of the control and remission groups, and were negatively correlated with the apoptosis of PMN and positively correlated with the disease. In the remission group, there was no significant difference between the above cytokines and the control group. Conclusion: The apoptosis of PMN in UC patients is delayed, and is closely related to the condition and curative effect. Excessive production of various inflammatory cytokines may be an important mechanism leading to delayed apoptosis of PMN
